There are several methods make PBS. Some methods not contain potassium while others contains calcium or magnesium. Most common method is given below.

10 L stock of 10 x PBS prepared by dissolving following ingredients-

800 g -NaCl (Sodium chloride)

20 g -KCl ( Potassium chloride)

144 g -Na2HPO4 · 2H2O (Disodium phosphate) (DSP)

24 g- KH2PO4 (Potassium dihydrogen phosphate)

8 L of distilled water.

After complete addition and mixing, make up to 10 L. The pH of the 10 X stock is around 6.8, but when it diluted to 1 x PBS it should be changes to 7.4. While preparing buffer solutions keep in mind that measure the pH using a pH meter. If needed pH should adjusted using HCl or NaOH.

Upon the dilution the resultant 1 x PBS is with final concentration of 137 mM NaCl, 10 mM Phosphate, 2.7 mM KCl, and a pH of 7.4.
