Collagen has a distinctive polypetide composition. One-third of its residues are Glycine and other 15-20% of its residues are Proline. After the polypeptide is synthesised, some of the proline residues are converted into proline derivatives.

The Proline residues cannot take up up tha alpha-helical backbone conformation (due to presence of a ring in their backbone) and lack the N-H groups that form the interhelical hydrogen bonding, thereby preventing collagen to assume a alpha-helix structure.
