We typically measure isotopic composition as ratios of isotopes because this ratio is constant. If we have different samples of a given element, then the number of atoms of each isotope will different in different samples. But the ratio of isotopes will be same in different samples. Also from mass spectroscopy, one can easily calculate this ratio.
