In this experiment to amplify regions of 16S genes, you are performing a PCR without added DNA? Why? If an amplified PCR product is detected in this reaction, describe an appropriate course of action for how you might proceed with your experiment.

  • The PCR amplification is performed using 16S rRNA. The 16S gene codes for 16S rRNA which is a component of bacterial ribosome. Hence DNA is not required as template for the PCR amplication (for this experiment).
  • The RNA is reverse transcribed and amplified.
  • Once the PCR product is detected, it is then sequenced using Sanger Sequencing.
  • Once we obtain the sequence information of the 16S rRNA, we can figure out the phylogenetic relatioship using the sequence information.
