  • SIMs teat is Sulphur Indole Motility Test.This test is done to determine the motility of bacteria.Motility is the ability of an organism by move by its own using motile structures like flagella or by fibrils.The medium used in SIMs test is Sulphur Indole Motility medium that is a differential medium that tests sulphur reduction,Indole protection and motility of bacteria.Motile organisms will spread into the medium from the point of inoculation.They will diffuse into the medium.While  non motile bacteria will remain at the point of inoculation.It never moves from the site of inoculation.Inoculation techniques are methods by which the bacteria is transferred into the culture medium.In SIMs test,the inoculum is stabbed into the center of a semisolid agar deep using a sterile inoculation needle.If there is a diffuse zone down the point of inoculation,it shows that the bacteria is motile.Inoculation technique is I important in  SIMs test because if the inoculum is placed in the medium in a streaked manner ,the movement of bacteria  cannot be identified.The inoculum should be placed in a single point in the medium.Then only we can determine whether the bacteria is motile or not.
  • SIMs test is usually done to differentiate members of enterobacteriaceae.They are gram negative bacteria.The staining technique used is gram staining.First the culture is stained with a primary stain called crystal violet.This gives a purple colour.When decolorized with ethanol and stained with counter stain safranin gives a pink or red.
