4d -
SET1 proteins have 1080 amino acids. So the number of nucleotides that are present in the gene will be 1080*3= 3240.
Length of the pRS406 plasmid is 4384 base pair. So total length of plasmid + insert = 3240+4384 = 7624 base pair.
It will little less than this as some bases will be removed during digestion of the plasmid with the restriction endonuclease.
4e - Bacterial cells have all the genes that are required for their growth. the pRS406 plasmid has an ampicillin resistance gene. If we grow the transformed cell in the presence of ampicillin, the cell will utilize this plasmid.
It has lac promoter which can be induced by IPTG and plasmid will start expressing the protein. But it may produce a functional protein or not is depend on the post-translational modification that is required for the activated protein.
