1. Here you did not provide total counted cell or number of cells present in ml or ul. How can I calculate with this incomplete information. For 1h, what is their cell density is also not given. It is quite difficult to calculate the number of cells present square with this insufficient data.

2. It would be ~0.53, but it depends on cell growth or death that you did not mention any conditions here. I have calculated for 1h that is ~0.066 and then calculated for 8h that it would be ~0.53 (0.066x8).
