DNA methylation is essential for gene regulation and cell differentiation but in a regulatory method. DNA methylation protects DNA to bind with replication or transcription proteins but demethylation is also important to cause differentiation of cells. Methylation and demethylation both are important for the survival of cells.

DNA methylation is important in mismatch repair of the new unmethylated strand. However, the overexpression of DNA methylase causes premature methylation. This situation brings mutations in newly synthesized DNA because the premature methylated strand cannot be repaired.
