The CRISPR-Cas9 gene editing system is a bacterial immune defence system. PAM sequences, when present adjacent to the target sequence, promote the cleavage of the complementary DNA strand by Cas9 after crRNA (or guide RNA in gene editing) recognizes the target sequence. Bacterial genomes do not have PAM sequences to avoid self-targeting by the CRISPR-Cas9 system.
