The purpose of doing negative staining is to know the structural and/or morphological features of the cells, bacteria, macromolecules, etc.

When samples are stressed, they tend to differ in their structural and morphological features compared to their normal condition. So, when someone uses the stressed sample in negative staining he/she will not be able to visualize the actual structural or morphological features of the target sample. So, one has to know if his/her sample is fresh or in stressed condition before doing negative staining.

That is why the negative stains should be done with a fresh sample rather than a stressed sample.
