Thermal death point (TDP) is the temperature which is required
to kill certain bacteria and is of two types i.e. lowest
temperature required to kill bacteria and highest temperature
required to kill bacteria when exposed to the respective
temperature for 10 minutes.
The following experiment can be designed.
1. Prepare culture suspensions in test tubes using Nutrient Agar
Broth.
2. Expose the broth solutions at different temperature for a
duration of 10 minutes. (Ideally from -20 degree Celsius to +20
degree celsius)
3. Prepare Nutrient agar plates and divide it into 4
quadrants.
4. Inoculate the culture suspensions and let it incubate at
37-degree celsius for 24 to 48 hours.
5. The highest temperature labelled quadrant which showed no growth
of culture indicated the TDP of the bacteria.
