I am tagging dihydrofolate reductase (DHFR) with GST, Myc-FLAG, and His. It is asked my hypothesis of whether they will appear at the N or C terminus and I do not know how to predict that. If someone could explain this, it would help a lot.

GST, Myc-FLAG, and His tag can be used to tag proteins of interest, for example, dihydrofolate reductase (DHFR) in this case. Typically, all of them are protein/ amino acid tag of different size. The choice is made on the basis of size of protein of interest as well as its compatiblity for the same. These tags are inroduced using recombinant DNA technology and hence can be tagged at both N- terminal or C-terminal. However, when tagging at N-terminal it should be incorporated after signal peptide sequence.
