Western blog is a immunological technique used to detect proteins. It is also called as Immunoblotting. Proteins are first subjected to gel electrophoresis, transferred to a membrane support and the protein of interest is detected with an antibody.

At first the protein extracted from the sample by subcellular fractionation or cell lysis.

Then the proteins are electrophorsed in agarose or poly acrylamide gel.

After electrophoresis the separated proteins are transferred to a membrane support as nitrocellulose or PVDF membrane.

The transfer of proteins from gel to membrane is carried out electrophoretically. The membrane placed on the anode side of the gel. An uniform electric field created across the thickness of the gel. Proteins migrate out of the gel and a replica formed on the membrane.

Then the membranes are washed with buffer to remove loosely bound proteins.

The membranes are then incubated with primary antibody. This step is little bit time consuming.

Then the membranes are washed to remove non-specifically bound antibody .

After that further incubation occurs with secondary antibody conjugated with an enzyme. These secondary antibody will bind the primary antibody.

The membranes are then incubated with substrates of the enzyme which will form insoluble coloured products.

So a coloured precipitate formed on the membrane in the position of target protein.
