The structure of intercalating anticancer agents including epirubicin comprises of 4 cyclic rings (tetracyclic ring system), which are six membered. They also contain one sugar substituent. The family of compounds to which such structures belong are called Anthracyclines.

The phosphate ion present on the backbone of DNA is negatively charged where as the ammonium ion present in the intercalating cancer agent is positively charged. Due to the opposite charges of the two ions, a strong ionic is formed between the two groups. Hence, the molecule gets locked in position.
