1- False negative results due to improper experiment. Here negative means the (absence of a condition) i.e. the presence of something is not there. But in reality, it is present. You can observe false positive results because of certain experimental inhibitors present in your samples which hinders the positive result and give negative results. Consider a situation of confirming the presence of a virus in your sample, but because your plasma contains RT PCR inhibitors you don't see the amplification of the viral gene. But its there.

2- In this case, we can do direct ELISA. It includes bind of the potato samples hypothetically contains virus onto the surface, followed by incubating with the antibody bound with the enzyme that will attach to the viral antigen on the samples over the surface. Then we can add the substrate that with react with the bound enzymes and give the signal. The occurrence of the signal will suggest the viral infection in the potatoes is positive.
