Temperature : Temperature changes can mess up the reaction and the enzyme won't work. High enough temperature will cause the enzyme to denature and have it's structure start to break up.

pH : Extreme pH value will cause enzyme to denature.

Heavy metals : heavy metals act as poisons to enzymatic activity. Heavy metals ions react with S-H group of cysteine bonds, forming a covalent bonds with sulphur atom and displacing the hydrozen ion. This causes the enzyme to lose its ability to catalize reaction.

These factors affect the enzyme activity.
