First, to visualize any organelle stain it with appropriate stain, janus green B for mitochondria, acetocarmine for nucleus etc. All of the organelles are scattered inside the cell, except for nucleus, which is one in one cell mainly.
To visualize organelles at various depth, you need to use coarse and fine adjustment knobs. When you use coarse adjustment know, stage moves in an upward or downward way. The distance between the objective lens and specimen (present on stage) changes. Therefore, it changes the focal plane of the specimen. Different focal planes have different type and number of organelles.
Now, you have located an organelle in the cell using coarse adjustment but it is not clearly visible to you. So you will now use fine focus knob to sharpen the image. It is increase detailing of the image. Most the internal structure can now be seen.
