6. The transposon Tn5 was used to create a library of 50,000 Escherichia coli mutant strains having random Tn5 insertions in their chromosomal genes. A suicide delivery plasmid vector that carries Tn5 was used for this purpose. Explain how and why a suicide delivery plasmid was used to generate this library and give one (1) example of how you would screen this library to find mutants with Tn5 insertions in specific genes of interest, for example, in the lac (lactose) operon. (10 marks).

Suicide delivery plasmid vector does not have ori site in its plasmid so plasmid not able to replicate in the bacterial cell.
After number of generation the plasmid started to dilute in the cell and it gets eliminated is refer as committing suicide. In Tn5 transposon the transposon are transfered and unwanted part of vector get diluted. In lac operon insertion the transpon Tn5 have same restriction enzyme sites in lac operon and same sequence present before and after of transposon construct. After this vector insert into bacterial cell transposon or jump gene transfer from insert in to lac operon.
