  • MacConkey Agar is a selelctive media for Gram negative bacteria, specifically Enterobacteriaceae.
  • This selectivity is facilitated by the presence of pancreatic digest of gelatin, proteoses peptones, mainly bile salts and crystal violet (which inhibit Gram positive bacteria)
  • MacConkey Agar also acts as differential media and differentiates lactose fermenting bacteria from non-fermenting bacteria.
  • Presence of lactose as carbohydrate source, help in such differentiation.
  • Organisms that can ferment lactose will produce lactic acid.
  • Neutral red is used as indicator, which is colorless at pH greater than 6.8 and turn red in lower pH of less than 6.8.
  • Thus, if lactose is fermented, acid produced causes change in pH.
  • Observation is as pink/red colonies for lactose fermenting bacteria.
