1. The sarcomere length decrease or contract in third test tube because glycerinated muscle contracted in presence of ATP and without ATP, activation of myosin head in muscle is not possible. Mg ions interfere with muscle contraction in case of live tissue but in glycerinated tissue , MgCl2 and KCl solution addition to ATP increases the muscle contraction due to mysosin's high compatibility to these ions.
