1. In spectrofluorometer transparent cuvette is used as the light path is different than uv-spectrometer. The light enters through one side and exits through the opposite side after passing through the sample (exciting fluorescence as it passes). This light is omitted .i.e. it is not collected by the detector. Fluorescent light that leaves the cuvette sideways, at 90 degrees to the excitation beam, is collected by a detector.

2. In flame photometry the species (metal ions) used in the spectrum are in the form of atom. Flame photometry is good only for elements that (Na, K, Li, Ca) are easily excited and do not require very high temperature. It is based on the measurement of intensity of lights emitted when the metal is introduced in the flame. Here the detection limits were quite high. Hence it does not require high resolution monochromator.
