Inoue transformation buffer consists of PIPES (piperazine-N,N′-bis(2-ethanesulfonic acid)) buffer pH 6.7, MnCl2, CaCl2 and KCl. Buffer is filter sterilized before use. The transformation protocol using Inoue transformation buffer was described in a research paper in 1990 (High efficiency transformation of Escherichia coli with plasmids, Inoue et al. 1990 Gene 96, 23-28). The the competent cells are prepared from conventional CaCl2 method that uses heat shock technique. In this method, the bacterial cells are grown at 18°C and ice-cold transformation buffer (TB) is used for preparation of competent cells.

Inoue transformation method is used to store cells in a competent state. The overall transformation efficiency is also improved using this technique.
