Gene A is essential for early embryonic development in human. A mutant version of A (Am) causes cell number to decease in human embryo during its early developmental stage. A is highly conserved between human and Drosophila, indicating the functional importance of A. It is known that the homozygous Am/Am Drosophila is embryonic lethal. You were tasked to design an experiment to study the question how Am causes cell death in Drosophila. What sequences should the completed chimeric plasmid have?

The gene A codes for growth substances which are essential for the cell growth and increase in number in embryonic stage. When A is mutated, it gets converted to a recessive allele, which in homozygous condition inhibits the growth of the cells. It leads to making the Am allele lethal.

Perform the experiment by self. Am allele on being homozygous condition, leads to abnormalities due to the lack of formation of essential components of growth. In heterozygous condition the Am allele fails to express, as it is compensated by the normal A allele.

So if want to insert external genes by recombinant DNA technology, then it requires the nitrogenous base sequences responsible for the production of desires factors.

Chimeric plasmids are the hybrid chromosome or DNA made up of genes of two different organisms. Here the chimeric plasmid must contain gene responsible for the normal growth of the embryo. The chimeric plasmid should contain the A gene. Chimeric plasmids can be inserted using the r-DNA techniques.
