Stem cells of meristematic region are maintained in an undifferentiated state by the signals of organising centre. There is a feedback signalling group present between the stem cells and organising centre, which balances stem cell renewal versus differentiation, which allows the plant to maintain the organization of the shoot meristem.
The presumed stem cell region is characterized by the expression of the CLAVATA3 (CLV3) gene, CLV3 gene is used as an operational stem cell marker for the shoot meristem.
The CLV3 gene encodes a small peptide. This peptide is secreted into the extracellular space. This peptide acts as a ligand for the CLV1 receptor-like kinase in the extracellular space. CLV1 receptor-like kinase is expressed throughout most of the shoot meristem.

SHEPHERD is an HSP90- is the protein that acts as the pacemaker. This is required for CLV signaling.
