Why acid-fast bacteria can resist decolorization by acids during laboratory staining?
Carbol fuchsin, which is the primary dye used in acid-fast staining, along with application of heat, penetrates the lipoidal cellular material present on the cell, and enters the cytoplasm, thereby staining it red. Now, the decolourizing stain, which is 3% HCl in 95% alcohol, enters only the cells which is non-acid-fast, that is, which has been stained by carbol fuchsin, and decolourizes it. Such a mechanism is not possible for acid-fast cells as those cells lack the lipoidal material. As a result of which it prevents the decolourizing stain from penetrating into the cell.
