Are the E. coli BL2I thermophiles? If not, explain how can it be transformed to be thermophiles (think about recombinant and gene expression methods)
Thermophilic Bacteria is the one which is able to live under high temperature condition even more than 50 degree Celsius. Mesophilic Bacteria is the one which is able to tolerate only normal temperature of around 20 to 37 degree Celsius.
We can transform a mesophytic bacteria into thermophilic bacteria using Recombinant DNA technology.
Procedure -
Isolate chromosomal DNA from thermophilic bacteria.
Using specific primers amplify the gene responsible for enabling the bacteria to live under high temperature condition.
Isolate plasmid DNA from mesophytic bacteria.
Using DNA ligase enzyme, join the amplified and plasmid to form a Recombinant DNA molecule.
Insert this Recombinant DNA molecule into the mesophytic bacteria for its stable integration and maintenance.
The mesophytic Bacteria is having the gene for enabling It to live under high temperature condition.

(c) An E. coli strain was created by transforming with the two plasmids shown below: pm Tet' gene pxyls HOC 5 G. E. coli Amp' gene Kan gene Mutants of this transformed E. coli strain can be obtained by exposing the cells to dimethyl sulfate. Briefly, explain how you might screen these mutants to obtain an xyls variant that can bind to 4-ethylbenzoic acid 1, carefully explaining the logic underlying your protocol. You can assume that wild type xyls cannot...
E. coli strain BL21(DE3) was transformed with the pET30a+ plasmid containing the ilvC gene that encodes for E. coli KARI. The starter culture was incubated overnight in LB broth containing 50 µg/mL kanamycin at 37°C. The sample were already added IPTG and the OD600 reaches the 0.6-0.7 range. How to improve the purity and yield ?
You set up a transformation hoping that a strain of E. coli will take up a plasmid containing an ampicillin resistance gene, making the strain able to grow on LB + amp. To see if the transformation worked, you set up four plates: (1) Untransformed E. coli cells on LB plate (2) Untransformed E. coli cells on LB + amp plate (3) Transformed E. coli cells on LB plate (4) Transformed E. coli cells on LB + amp plate. Your...

Which of the following is considered as a vector? GFP gene C E. coli cell c ampicillin-treated agar plate 0.5 points QUESTION 2 1. What is the function of bla gene? Break down the ampicillin Regulate the transcription of GFP gene Express the fluorescent trait in bacteria Control the uptake of DNA molecules in bacteria...
Expression of a heterologous protein in CHO cells and in an E. coli system produced proteins with the same molecular weight on SDS-PAGE. Subsequent in vitro bioactivity studies demonstrated high-level activity with the CHO cell-derived recombinant protein, but no bioactivity with the E. coli derived product. Explain this discrepancy, and propose an experiment to test your hypothesis

Explain how you might engineer E. coli to produce human growth hormone (HGH) using the following. E. coli containing a plasmid DNA carrying the gene for HGH DNAIigase A restriction enzyme Equipment for manipulating and growing bacteria A method for extracting and purifying the hormone An appropriate DNA probe For this problem, assume the HGH gene lacks introns.
Dr A. Rachnid plans to express the gene for spider silk in E. coli(to make her fortune). The gene has three exons. Draw the spider silk gene, labelling introns and exons and showing where both transcription and translation start and stop. (3 marks) Explain how Dr Rachnid should modify the spider silk gene so that it can be expressed in E. coli. (2 marks) She makes the modifications and all seems to be going well until several workers in her...
