Absence of isoamyl alcohol in the Kovac's reagent while testing of an indole positve bacterium will produce an overall cherry red coloured solution, instead of a cherry red colored ring.

The isoamyl alcohol forms a complex with rosindole dye, which causes it to precipitate. Because isoamyl alcohol is not water soluble, the cherry red color ring will form at the top of the broth.
