There is need for visualizing various organisms in different microscopes as they differ in level of organization.

1. bright field microscope -

  1. used to observe the morphology of microorganisms such as protozoa, bacteria, fungi in living ( Unstained) & non - living ( Strained state )
  2. x1000 magnification
  3. can observe <0.2 micro meter diameter thick eg: spirochetes , virus.

2. darkfield microscope -

  1. Unstained organism against dark background
  2. used for thin spirochetes

3. Phase contrast microscopy

  1. resolving power - 0.2000 micro meter
  2. can be used to see unstrained organisms
  3. magnification - x1000

4. Polarising light microscopy

1. contrast-enhancing to evaluate composition

2. 3 D structure of anisotropic specimens.

3. It has polarising filters to make use of polarised light.
