Answer #1

Smears are prepared for the study of the bacterial culture. The purpose of this process is that after it the smears will be stained so the bacterial colony can be viewed. Without staining the colorless bacterial colony is not visible. But to stain it properly it requires to be in the smear form.

The smears are thin layers of the colony which can be fixed on the slide easily. Smears are prepared using four methods which are water wash, blood film, drop and rest, and one conventional method.

The smear making from the solid medium needs some drop of water to be added but the smears being prepared from the liquid medium don't need elwater to be added.
