Human Nucleosome is made of histone proteins ocatamer in humans and DNA about 200 bp . Linker DNA (about 80bp) between two nucleosomes is protected by one type of histone H1. So when we carry out digestion by nuclease enzyme which cut only naked DNA not protein coated DNA we get different sized DNA fragments .Histone H1 link nucleosomes together and compact chromatin in higher level structures like 30nm fibre.

Case (a) If Nucleosome is absent then many small DNA fragments of less than 200bp are obtained in well no 1 of gel giving thick DNA band.

Case (b)If loose protein like linkers H1 are absent then chromatin assembly or compaction will be difficult and only two bands representing core DNA 200bp and linker DNA about 80 bp are obtained.

Case (c)Large Nucleosome (400bp ) along with linker histone will give three band pattern in gel as assembly of Nucleosome into higher level chromatin will be possible (80bp, 400bp, 800bp ......multiples of 400)

Case(d) Human like Nucleosome pattern will correspond to pattern of band in last well no. 5 as linker DNA band of about ,80bp and multiples of 200bp can be seen. This also represent compaction of DNA into higher level of structures .

Case (e).Huge Nucleosome of about 600bp and linker protein will give band pattern as in well no 2(80bp , 600bp , 1200bp and so on ) due to possibility of condensation process via human like linker protein histone .
