In presence of 5% NaOH, benzocaine undergo Hydrolysis to give p-aminobenzoic acid which become soluble in water. Thus, after addition of NaOH, benzocaine get Hydrolyzed and go in the aqueous layer hence organic layer is free from benzocaine.
