  • Mutation in Rb gene prevents the production of tumor suppressor protein of the gene product called pRb.
  • The non-phosphorylated form of pRb prevent progression of cell cycle beyond S-phase.
  • Without pRb uncontrolled cell growth results in tumor formation.
  • Upon phosphorylation, the transcription factor E2F is released from RB and help in transcription. This results in transition of cell cycle from G1 to S phase.
  • Upon mutation, E2F is constitutively released, resulting in uncontrolled cell division.
  • RB-E2F association is mediated though phosphorylation of several amino acids (like alanine).
  • If there is mutation in the phosphorylation sites, this leads to conformational change and release of E2F.
