Phenol red fermentation test.

Phenol red broth is a differntial test medium used to differentiate gram negative enteric bacteria.It contains peptone,phenol red which is the pH indicator,a durham tube,and one carbohydrate,either glucose,lactose or sucrose.This carbohydrate fermentation test is based on to determine whether or not a bacteria can utilize a certain carbohydrate.If tests for the presence of acid or gas produced from carbohydrate fermentation.The media in each tube contains a carbohydrate.A color change from red-orange to yellow occurs when acid is produced as a result of carbohydrate fermentation.A durham tube may be inserted in a tube of phenol red broth w/ carbohydrate to allow for the detection of gas production.Lack of yellow color is the indicative of a negative carbohydrate fermentation.Gas formation is indicated by the appearence of gas bubbles in the durham tube.
