How can Chimeric proteins be used to study protein sorting?

Chimeric proteins are translated from the RNA which is comprised of exons of two or more different genes. When a chimeric protein has a Glutathione S-transferase (GST) tag or a hexa-his peptide e.t.c, they can be used for protein sorting by using affinity chromatography with glutathione immobilized to a solid support (glutathione is the substrate for GST) or nickel or cobalt resins (histidine residues binds to immobilized metal ions, nickel, cobalt).

What effect would the addition of a lysosome‐targeting signal have on the subcellular localization of  

a. A protein that is normally cytosolic?

The normally cytosolic protein does not have a signal sequence and does not enter the endoplasmic reticulam. Hence, addition of a lysosome-targeting signal would have no effect.

b. A protein that is normally secreted?

The addition of lysosome-targeting signal  would direct a normally secreted protein to lysosomes from the Golgi apparatus.
