A. Secondary structure not really favours hybridization. Instead dehybridization is favoured by it. Hybridization in the other hand is favoured by the primary structure. Regarding absorption, single stranded helix are much more prone to get absorbed on the gold nanoparticles.
B. A260/A280 ratio tells about the purity of the DNA molecule examined. Thus radio frequency ablation will defenitely affect this ratio which is strictly a purity measure.
