Paper Chromatography: Why does spotting the known and the unknown on the same paper help identify the unknown amino acid? Why does spotting both the known and the unknown in the same place on the paper help to identify the unknown amino acid?

Paper chromatography helps to identify unknown compounds. If you spot the known and unknown amino acids in the same paper and you add mobile phase , both compounds will travel to certain distance. If both have travelled same distance from the spot, then you can say that known and unknown are same amino acids.

Similarly, if you spot both unknown and known amino acids on the same place and add mobile phase, both known and unknown compounds travel same distance and form single spot this indicates known and unknown are same.

If known and unknown are different, then it gives two different spots after adding the mobile phase.
