1. INTRODUCTION

the respiratory disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) firstly reported in China, from there it was spread to all over the world, it is a positive-sense single-stranded RNA virus,   and spread in humans. SARS-CoV-2 is closely genetically similar to coronaviruses of the bat, An intermediate animal transmitter is a pangolin who is responsible for the introduction in humans. we can Real-time reverse transcriptase–
polymerase chain reaction (RRT-PCR) for the detection of the virus.

2. OBJECTIVE

isolation of SARS-CoV-2 in specimens of Different patients

3. METHODOLOGY

a. you can investigate the distribution of SARS-CoV-2 among tissues of various patients infected with coronavirus, diagnosis based on symptoms and confirmed by SARS-CoV-2 detection. patients samples can be obtained from hospitals.

b. you have to collect Pharyngeal swabs from previously admitted patients, you can also take samples of blood, sputum, feces, urine, and nasal sample.

c. collect extracted RNA from clinical specimens and determine that sequences by the help of
rRT-PCR

d. The cycle threshold values of rRT-PCR will be used as indicators of the copy
number of SARS-CoV-2

e. that RNA in specimens will have lower cycle threshold values that are less than 40 is considered as positive for SARS-CoV-2 RNA.

f. you can culture SARS-CoV-2 positive specimens and then isolate microorganisms.
e. electron microscopy will be performed for the detection of live virus.

4.EXPECTED RESULTS

Themeancycle threshold values less than 40 is considered as positive for SARS-CoV-2 RNA. of all specimen types.

5.ALTERNATIVE STRATEGY

cell culturing of infected tissue then study the sequence of cultured tissue and, match observed sequences of the genome with SARS-COV 2

In this study, SARS-CoV-2 will be detected in specimens from multiple sites of different numbers of patients with COVID-19. The important point to be noted that the live virus will be detected in feces, implying that SARS-CoV-2 and may be transmitted by the fecal route. so results completely depend on the methodology and definitely it will be successful.
