Why, the cooperative action of both enhancers stimulating much stronger expression than just that of enhancer 2?

Enhancer-enhancer pairs had higher expression correlations than non-interacting pairs of enhancer.

The identified enhancer-enhancer interaction networks present highly dynamic linkages. Cooperative actions of enhancers increase the transcription than that of the action of just enhancer 2 alone.

What are enhancers doing?

An enhancer is a short region of DNA that contains 50–1500 bp, bound by activators to increase the transcription of a particular gene will occur.

They are cis-acting, regulatory binding sequences or sites of DNA for transcription factors that enhance the transcription of an associated gene by a activating promoter, thereby "turn on" the gene expression.

What is mediator?

Mediator act as transcriptional coregulator that binds to an activator to increase the transcription rate of a eukaryotic genes. This multiprotein complex interact with transcription factors and RNA polymerase II to transmit signals from the transcription factors to the polymerase.

Mediators involved in signal transduction, chromatin organization, RNA synthesis, diseases etc.
