Do antibodies have the capability to recognize amino acids when they are phosphorylated?

If so, describe an experiment using Immunohistochemistry that would allow you to determine if a specific serine/threonine residue has been phosphorylated.

Yes antibodies has capability to recognize a specific phosphorylated amino acid in a protein sequence..

For this we have to use anti body agsinst particular phosphorylated amino acid in the experiment. These phosphorylated antibodies are also known as state specific antibodies, which recognizes specific states of protein .

Immunohistochemistry is a process which selectively identifies presence of a specific protein in cell by using principle of antigen antibody interactions. Steps used in IHC are following.

1. Cur thin sections of tissue on the coated slides .

2) removal of paraffin, and dehydration of tissue by increasing radiation of alcohol treatment.

3) antigen retrieval by using appropriate buffer example citrate buffer

4) blocking with serum

5) add primary antibody appropriate concentration ( phosphorylated antibody) incubate for overnight at 4 degrees.

6) next day 2ash properly and add secondary antibody ( antibody against primary antibody). Again si proper washing.

7) add substrate in to slide .enzyme linked with secondary antibody react and produce born color.

8) again cause dehydration and paraffin treatment and ad counter stain and mount the slide.

Where brown staning present in cell show presence of phosphorylated protein in cell.
