Increasing proton leakage through an uncoupling protein would tend to decrease the rate of superoxide production. The proton leakage through UCP1 leads to phosphorylation of ADP to ATP which increases oxygen consumption and heat production in the mitochondria of brown adipose tissue. This happens due to increased proton conductance of the mitochondrial inner membrane because of the presence of huge concentrations of UCP1. UCP2 and UCP 3 are present in lower concentrations due to which the uncoupling associated with them has little effect on heat production. The proton conductance is increased by superoxide through effects on UCP1, UCP2, and UCP3. Purine nucleotides inhibit superoxide induced uncoupling and proton conductance which is overcome by the release of fatty acids from intracellular triacylglycerol as a result of adrenergic activation. Adrenergic activation takes place in response to cold.
