Suppose that you would like to use hollow-fiber dialysis cartridges for engineering large portions of cortical bone that mimic the native structure of this tissue. Such a cartridge has many "fibers'' running from one end to the other, where the fiber wall functions as a semi-permeable membrane, allowing solutes up to a certain size to diffuse through. By flowing a nutrient fluid through the fibers, it is possible to achieve substantial transport to (and from) the material within the overall large dialysis chamber, and between the fibers. You are able to seed the cartridges (outside the fibers) with a material that includes cells and cortical bone-like matrix. Your strategy is to use the hollow fibers to provide nutrients and remove waste products from the tissue, via diffusion. Assume that the flow rate is fast enough that there is little change in the concentration your molecule of interest from inlet to outlet. How would you design such a system? Issues to address include how many cells are typically in a region of bone, how much oxygen and nutrients that the cells metabolize and at what rate, what nutrients and regulatory factors to deliver to the bone cells in the nutrient media, what membrane pore size is appropriate, how many fibers and what size fibers per cross-sectional area.
