Explain what a volume-to-volume dilution refers and describe in sequential order, how would one make a 1:10 dilution of 10X TBE buffer using deionized water as your diluent. Show any calculations.

First Part:

Volume to volume dilution: It refers to the ratio of a solute to the final volume of the diluted solution. For example, if you wanted to make 1:100 dilution of a particular solution you would mix 1 part of that solution with 99 parts of diluent, i.e., 1 mL of solution with 99 mL of diluent if final volume is 100 mL.

Second Part:

Given, dilution ratio = 1:10, Initial concentration of TBE buffer = 10 X

So, to make 1:10 dilution of 10 X TBE buffer we will mix 1 part of 10X TBE buffer with 9 parts of deionized water. For example, 1 mL of 10 X TBE buffer with 9 mL of deionized water if final volume is 10 mL. Final concentration of TBE buffer will be 1 X (Calculated by using formula V1 C1 = V2 C2).
