The active site of enzyme is lined by specific amino acids. All these amino acids interact with each other to give a definite shape to the enzyme active site. All these amino acids have specific sidechain which interact with the substrate molecule and convert it to the product. Any change in the amino acids present in the active site only to the change in shape of the active site of enzyme, thereby changing the specificity of enzymes for that particular substrate. In this case the substrate would be able to bind to enzyme and no catalysis would occur.
