The amino acid Prolinehas a pk1 of 1.99, pk2 of 10.60. The amino acid Serine has pk1 of 2.21 and a of pk2 of 9.15. If these two amino acids bind together by peptide bond creating the peptide sequence Pro-Ser . What will be the overall charge of this peptide at the physiological pH of 7?

Answer #1

At pH = pK half of the dipeptides (Pro-Ser) are deprotonated.

When pH >> pK, then all of the dipeptides are deprotonated.

At pH = 0, both the amine and the carboxylic acid groups will be protonated. Thus, the overall charge on the dipeptide at this pH = +1.

At pH = 7, the amine group will remain protonated but the carboxylic acid groups will be deprotonated.

Therefore, the overall charge of this peptide at the physiological pH of 7 = 0
