As the previous protocol being used is not given, we can assume that using ferric chloride in the media will help in detecting if the genes for esculinase activity are mutated or not.

If the genes are not mutated, then esculinase activity will take place to hydrolyze Esculin into esculetin and dextrose. The esculetin will react with ferric chloride and form a black brown colour. So, if the black brown colour is not formed, then we can suggest that the genes for esculinase activity are mutated.
