In this case, we just need to observe the culture under the microscope. Remember that eukaryote cells has a defined nucleus which means that if we observe the culture under a microscope we should observe the cell and the nucleus (under a 40x objective). If we found the nucleus, we can conclude that our culture is formed by eukaryotic cells. On the other hand, eubacteria is a gram-positive genus which means that if we take a small sample of the culture and we make a gram staining assay, we should observe gram positive bacteria under the microscope (after staining). If we don’t get a positive result from the staining, we can conclude that our culture is formed by Archaea.
