Ans: The volume of DNA solution was 0.5 mL.

(a) DNA concentration (µg/ml) = OD of DNA at 260nm  × dilution factor × 50µg/ml

= 0.459 x 1/100 x 50

= 0.2295 µg/ml

DNA yield (µg) = DNA concentration × total sample volume (ml)

= 0.2295 x 0.5

= 0.1147 µg

(b)  DNA concentration (µg/ml) = OD of DNA at 260nm  × dilution factor × 50µg/ml

= 0.167 x 1/100 x 50

= 0.0835 µg/ml

DNA yield (µg) = DNA concentration × total sample volume (ml)

= 0.0835 x 0.5

= 0.0417 µg
