A point mutation in position LMNA gene is the cause of progeria. Basically the point mutation in LMNA gene replaces a cytosine with thymine. This mutation results in creation of a 5' cryptic splice site within exon 11, that ultimately results in the production of shorter than normal mRNA transcript.

The translation of the shorter mRNA produces an abnormal variant of the prelamin A protein which is referred as progerin which cannot become incorporated as a structural part of the nuclear lamina.

The nuclear lamina lacking lamin A protein, could not be able to provide the nuclear envelope with enough structural support, and resulting it to take on an abnormal shape.
