AIM
To detect the presence and amount of acetobacter in wine.
METHOD
Acetobacter strains oxidised ethanol to acetic acid and forms carbon dioxide and water. Acetobacter is a gram-negative bacterium and are able to ferment glucose , lactose , maltose ,raffinose, galactose etc. The steps includes for detecting the presence and amount of acetobacter in wine are following ;
. Cultivate Acetobacter aceti on GYC agar at 30 degree celsius for 48 hours.
. Detecting the presence of acetobacter in wine , positive acetobacter is control an amplification plot and melting curve.
. DNA extracted from wine samples incubated for 1 week at 27 degree celsius aerobically.
. The susceptibility of acetobacter varied in different isolates from 102 - 107 CFU ml-1 .
RESULT
If the test is successful, the result is positive . That is acetobacter can be identified and the number of cells can be determined.
