In competitive inhibition, the inhibition constant (Ki) is referred as a dissociation constant of EI-complex, Ki=[E][I]/[EI]. Ki refers the potency of an inhibitor, it is independent of substrate concentration. The competitive inhibitor [I] increases the Km for enzyme without affecting Vmax. Larger Ki represents the lower binding affinity, results in the requirement of a larger quantity of inhibitor for competing with the substrate to bind with a respective enzyme.
